AN IMMUNOHISTOCHEMICAL STUDY OF THE EFFECTS OF CYCLOPHOSPHAMIDE ON THE DNA SYNTHESIS OF DEVELOPING RAT TOOTH GERMS

Abstract
-Abstract-
The purpose of this study was to clarify the effects of cyclophosphamide on the tooth
germs of developing rat mandibular lower incisors. Experimental animals were treated
with peritoneal injection of bromodeoxyuridine 1, 3, 7, 15 hours 1, 3, 5, 7, 11, 14, 21
days after peritoneal dine. They were fixed in 2.5£¥ glutaraldehyde through vascular
perfusion and their removed mandibules were decalcified with 10£¥ EDTA. They were
routinely processed for 4§­ paraffin actions, stained with H£¦E and were
immunohistochemically stained with anti-bromodeoxyuridine antibody by Avidin-Biotin
complex method. Bromodeoxyuridine positive cells were counted from area to area under
the light microscope.
The obtained results were as follows :
1. Gross examination showed that there were hemorrhage and the interrupted formation
of dentin and enamel on the 5th and the 7th days after peritoneal injection of
cyclophosphamide. And cavities were formed above basal portion of tooth germ on the
14th days after experiment, and regeneration of dentin, and enamel were seen in basal
portion on the 21th days after experiment.
2. Histopathologic examination showed dysplasia of undifferentiated mesenchymal cells
on the 1st day after cyclophosphamide injection, mitosis and hyperchromatism of
undifferentiated mesenchymal cells on 3rd day, acellular area as a result of destruction
of undifferentiated mesenchymal cells on the 5th day, and formation of osteodentin
inpulp on the 7th day, and regeneration of undifferentiated mesenchymal cells,
predontoblasts, and preameloblasts on the 21th day.
3. Electromicroscopical examination showed degeneration of euchromatin and
heterochromatin of undifferentiated mesenchymal cells on the 1st day after
cyclophosphamide injection, degeneration of nucleoli on the 3rd day, and regeneration of
components of nucleus on the 11th day.
4. Immunohistochemical examination showed abruptly decreased DNA synthesis of
undifferentiated mesenchymal cells, preodontoblasts, and preameloblasts from 1 hour to
5days after cyclophosphamide peritoneal injection.
5. Abruptly increased DNA synthesis of predontoblasts, and preameloblasts on the 11th
day of cyclophosphamide injection and that of undifferentiated mesenchymal cells on the
14th day presented regeneration of tooth germ.
